Dimitrescu Castle
You will be forever locked from Dimitrescu Castle if you collect the death flower dagger from the coffin after completing the Hall of Four Puzzles. Although you should go through Dimitrescu Castle and collect the 4 Angel Masks, entering the door and picking up the dagger will trigger a boss fight and you will never be able to return to the castle.
Dimitrescu Castle is a great area to explore, full of all sorts of things worth wearing. Therefore, you should collect the masks and complete the puzzle, but never pick up the death flower dagger until you are ready. If you open the coffin, you can spawn Lady Dimitrescu, which is great for exploration, so keep that in mind as you play.
House of Beneviento
After escaping from a terrifying giant baby in a doll workshop, you'll find yourself in a deadly hide-and-seek encounter with Angie. Once Angie is defeated and you grab the bottle, the door to the exit will open. However, if you leave House Beneviento, you can never come back.
That's not a big deal, because there's really nothing in House Beneviento worth sticking around for. That is, if you don't like the comfort of creepy dolls. Make sure you pick Angie up off the floor before you leave, as she is a valuable item that can be sold to Duke.
Moreau Dam
Moreau's Reservoir is an outlier because while players can explore most areas as they wish before moving on, Moreau's Reservoir is an area where there is a time limit on certain items. This is due to the fact that in some areas you are being chased by a giant lake monster and parts become inaccessible shortly after and during them.
Moreau's Reservoir really changes once you complete the Sluice Gate Puzzleas the water level drains away allowing you access to new areas. However, there is a Goat of Warding and File that can't be missed in this area, so try not to go too fast because you'll be locked in forever using the elevator to get back to the surface.
Village
After completing the Stronghold part of the game, it is vital that you do everything you need to in the village. This will be your last visit to the village, so any collectibles you're missing in this large area now is the time to do so or you'll have to wait for NG+ or reload an old save.
There's plenty to do in the village, from an assortment of meat to gather, Goats of Warding to shoot, treasures to hunt and Files to read. Just make sure you do it all before or after the Stronghold, like boarding Heisenberg's Factory will permanently lock you out of the village.
The Heisenberg Factory
Heisenberg's factory is a vast labyrinth full of loot and collectibles worth finding. However, Heisenberg's factory will be inaccessible once you progress to the control room after defeating Sturm in B1. As such, you should make sure to avoid B1 with Heisenberg's Wrench and instead use it at lower levels for loot.
Only when you are ready to continue and never return to Heisenberg's Factory should you use the Heisenberg Key in B1 and fight Sturm to continue. Remember, this is the final point of no return, so whatever you have left to do, it's time to do it before the end begins.
