The Resident Evil series has been known for featuring female characters in leading roles since the first game in 1996. Heroines like Jill Valentine and Claire Redfield complement such characters as Ada Wong, Lady Dimitrescu and others. It's interesting to look back and see the roles these characters played in their stories.
Some heroes and villains have remained memorable years after their debuts. This is mainly because of the performances they did and the roles they played in interacting with the rest of the cast. With over 25 years of performances, it's worth taking a look at the female characters who have influenced fans the most.

Alex Wesker

Alex isn't the most famous Wesker in the series, as Albert has his groove in that department. Still, she's a great antagonist from Revelations 2. What makes her interesting are her unique and somewhat understandable motivations. Most villains in the RE universe want world domination, ultimate power, or something along those lines.

The main thing Alex wants is a new body to transfer her consciousness into, as her original body is deteriorating due to illness. While you can see why she'd want a new body, the fact that she's kidnapped people and tried to take their bodies against their will means she's still very much a villain.

Moira Burton

Despite being Barry Burton's daughter, Moira is a bit under the radar as she only appears in one play and it's not a major part. That said, he's an interesting character, especially for the Resident Evil series.
Most RE protagonists, male or female, are either competent killers or will become so very quickly. However, Moira refuses to use the gun due to a traumatic childhood moment when she accidentally shot her sister. This gave her an intense fear of firearms. It's fascinating to see how she deals with this in Revelations 2, when she's put in situations where the weapon would be very useful.

Annette Birkin

Over the course of Resident Evil 2, Annette Birkin emerges as a complex character who is ultimately on the side of good but doesn't always do the right thing. This can be seen in her relationship with her daughter Sherry.
In RE2, Sherry is still a child, but Annette is too preoccupied with her work to be there for her child. In fact, even when the outbreak breaks out and her husband William becomes a rampaging monster, she still focuses on her research instead of trying to care for her child. Therefore, the woman can seem a little cold at times. However, she eventually proves that she cares about Sherry and feels bad for the mother she was. The character's moral gray is what makes her fascinating.

Helena Harper
Since Resident Evil 6 features multiple co-op campaigns, Capcom had to introduce a few new faces to fill the protagonist slots. One of those faces belongs to Helena Harper, who ends up being one of the most interesting playable characters in recent series history. She is a seasoned secret service agent who turns out to be a bit of a badass during RE6.
What makes her a truly compelling character is her story. Although she is generally a morally good person, she agrees to participate in an assassination to save her sister who is being held captive. This made her feel very guilty, something she struggles with for the rest of the play.

Mia Winters

Although not the main protagonist, Mia plays a huge role in both Resident Evil 7 and Village. A lot happens to her during those games. After all, in RE7, the virus keeps turning her into a monster, leading to brutal fights with her husband Ethan. She is then secretly locked up in the Village while Mother Miranda takes her place.
So even though he doesn't have that many scenes, the ones he does have are very memorable. However, she is a difficult character to judge because most of what makes her interesting is what happens to her rather than her actual personality. That said, he's still a solid character.

Ashley Graham

Before the release of the remake, it would have been impossible to imagine Ashley being considered one of Resident Evil's best sidekicks. The updated version of the character is much more competent and has an empathetic personality that accompanies Leon on his mission.
Ashley even saves Leon from several tight spots, justifying her position as his partner. While she never achieves main character status, her role keeps Resident Evil 4 from becoming a generic action story. Ashley doesn't get the most personality development, but that's understandable since she's a supporting character.

Ingrid Hunnigan

For someone who isn't even a playable Resident Evil character, Hunnigan sure leaves a mark. As Leon's main character, Hunnigan provides him with valuable information in both Resident Evil 4 and Resident Evil 6. Hunnigan's professionalism deserves recognition as he does not tolerate anything outside the scope of his job.
Despite being rarely seen in person, Hunnigan feels like a full-fledged protagonist because she shows up so often to guide Leona. Sure, it would have been better if Hunnigan took a more active role in this area, but her character's purpose is to provide fans with an interpretation of important parts of the story.

Sheva Alomar
Resident Evil 5 is one of the most action-packed installments in the series so far, so Chris needed a sidekick who could pack a punch just like him. Sheva brings the necessary physicality and can cut through enemies on her own. She will never run out of stamina or strength to take down Albert Wesker.
The downside to such a solid partner is that fans don't get to see much of Sheva's personal arc. But he also has the distinction of having a major role in defeating the series' greatest villain. It's a shame the series hasn't brought her back since her debut because Sheva has the screen presence to stand out from the crowd.

Rosemary Winters

Rose is perfectly positioned as a potential series leader going forward. As the main characters age, 16-year-old Rose can take the story into the future. Rather than being overpowered by her powers, Rose is shown to be a layered individual struggling to feel human.
Resident Evil only scratched the surface with her, leaving a lot for fans to fully connect with Rose. Still, her arc of coping with the loss of her father and learning more about the different forms of viruses is compelling. It will be interesting to see if her story continues to develop so she can become the hero Chris intends.

Lady Dimitrescu took the world by storm when she debuted in Resident Evil Village. Her theatrics and blunt approach is perfectly suited to a horror game as it ratchets up the pressure. Lady Dimitrescu has a richer background compared to many other villains, so there is more to explore about the character.
Her towering height and unique style add to her memorable character, though Lady Dimitrescu ultimately declines like the other antagonists. The series went for a one-off hit, given the character's impact, and there's little chance you'll see her again. But she did more than enough to connect with the audience by being entertainingly sly.
